Overview

Zejula + carboplatin + paclitaxel is a multi‑agent antineoplastic regimen combining niraparib, an oral poly(ADP‑ribose) polymerase (PARP) inhibitor marketed as Zejula, with the platinum agent carboplatin and the taxane paclitaxel, both standard intravenous chemotherapies. Niraparib selectively inhibits PARP‑1 and PARP‑2, blocking repair of single‑strand DNA breaks and inducing synthetic lethality particularly in tumors with homologous recombination deficiency, while carboplatin forms DNA intra‑ and inter‑strand crosslinks and paclitaxel stabilizes microtubules to arrest mitosis.[1][2][12][13] This combination is being investigated primarily in gynecologic malignancies such as ovarian and endometrial cancers, typically in contexts where carboplatin–paclitaxel serves as the chemotherapy backbone and niraparib is layered on to enhance DNA‑damage–driven tumor cell kill and provide maintenance benefit.[1][7][11][13]
