Overview

This is a combination regimen of three drugs: **zibotentan**, a selective endothelin A receptor (ETAR) antagonist; **carboplatin**, a platinum-based chemotherapeutic that causes DNA crosslinking and apoptosis; and **paclitaxel (Taxol)**, a microtubule-stabilizing agent that inhibits cell division. Zibotentan blocks ETAR-mediated signaling, which is implicated in tumor proliferation, survival, angiogenesis, and invasion, thereby potentiating the effects of cytotoxic agents. This combination has been evaluated primarily in the context of ovarian cancer, aiming to enhance antitumor efficacy compared to chemotherapy alone. Preclinical models and randomized phase II clinical studies have tested this combination to determine its effect on tumor response and progression-free survival in platinum-sensitive relapsed ovarian cancer[1][3][5].
