Zidovudine + acyclovir is a combination of two small molecule antiviral drugs. Zidovudine is primarily a nucleoside reverse transcriptase inhibitor (NRTI) used to treat HIV infection, functioning as a chain terminator in viral DNA synthesis by targeting HIV reverse transcriptase[4][5]. Acyclovir is a guanosine analog antiviral mainly active against herpes viruses such as herpes simplex virus (HSV) and varicella-zoster virus (VZV), inhibiting viral DNA polymerase after phosphorylation by viral thymidine kinase[7]. The combination has been studied for patients co-infected with HIV and HSV-2, where both drugs may be used simultaneously to suppress symptoms of both viruses; in such regimens, close monitoring for pharmacokinetic changes is advised, as the combination can alter individual drug metabolism and exposure, affecting maternal, fetal, and amniotic fluid concentrations[2][6]. There is no evidence of a fixed-dose branded combination product of zidovudine + acyclovir; administration typically involves separate drugs.
