Overview

Zidovudine + lamivudine + efavirenz is a fixed-dose antiretroviral combination used in the treatment of HIV-1 infection. This regimen combines two nucleoside reverse transcriptase inhibitors (NRTIs)—zidovudine and lamivudine—with one non-nucleoside reverse transcriptase inhibitor (NNRTI)—efavirenz. The NRTIs act by incorporating themselves into viral DNA and causing chain termination during reverse transcription, while efavirenz binds directly to reverse transcriptase and inhibits its activity through a different mechanism. Together, these agents suppress HIV replication by targeting the viral enzyme essential for converting viral RNA into DNA[1][4][6]. This combination has demonstrated greater antiviral activity and better tolerability compared to some regimens containing protease inhibitors[1]. It is indicated as part of highly active antiretroviral therapy (HAART) in adults with HIV-1 infection.
