Overview

Ziftomenib + quizartinib is an all-oral combination therapy currently being evaluated in a Phase 1 clinical trial for the treatment of relapsed or refractory acute myeloid leukemia (AML) and myeloid mixed-phenotype acute leukemia (MPAL). Ziftomenib (Komzifti) is a potent, highly selective small molecule inhibitor of the menin-KMT2A (MLL) protein-protein interaction, which is a key epigenetic driver in leukemias harboring NPM1 mutations or KMT2A rearrangements. Quizartinib (Vanflyta) is a selective second-generation type II FLT3 inhibitor that targets proliferative signaling. This combination, primarily investigated in a study led by M.D. Anderson Cancer Center, aims to provide a synergistic therapeutic effect by simultaneously disrupting epigenetic dysregulation and kinase-driven growth in patients with NPM1, KMT2A, or NUP98 mutations.
