Overview

Zilovertamab + ibrutinib is a combination therapy currently in clinical development for hematologic malignancies, notably mantle cell lymphoma (MCL), chronic lymphocytic leukemia (CLL), and marginal zone lymphoma (MZL)[1][2][3][4][5][6]. Zilovertamab is a fully humanized monoclonal antibody targeting ROR1, an onco-embryonic receptor tyrosine kinase-like orphan receptor 1, and inhibits signaling that contributes to tumor cell growth, survival, and proliferation[1][2][3][5]. Ibrutinib is a small molecule inhibitor of Bruton’s tyrosine kinase (BTK), a key enzyme in B cell receptor signaling[1][2]. The combination shows synergistic activity, with zilovertamab potentially overcoming mechanisms of resistance to BTK inhibition and improving anti-tumor effects in preclinical and clinical studies. The combination is being evaluated in multiple phase 1/2 trials and the global phase 3 ZILO-301 trial in relapsed/refractory MCL[1][3][5][6].
