Overview

**Zimberelimab + chemotherapy** is a combination therapy being investigated primarily for cancer treatment, particularly in advanced gastric and gastroesophageal junction adenocarcinoma with elevated AFP and negative HER2 status. Zimberelimab (GLS-010) is a fully human monoclonal IgG4 antibody directed against the PD-1 receptor, functioning as an immune checkpoint inhibitor to restore T-cell activity and promote anti-tumor immunity. As a combination, it is typically administered with standard chemotherapeutic regimens such as XELOX (oxaliplatin + capecitabine) and is also studied with agents like lenvatinib, a multi-tyrosine kinase inhibitor. The primary mechanism of zimberelimab is blockade of PD-1, preventing its interaction with PD-L1/2, thus reversing T-cell suppression in the tumor microenvironment.
