Overview

Zimberelimab + domvanalimab is a combination of two investigational monoclonal antibodies developed for cancer immunotherapy. Zimberelimab is an anti-programmed cell death protein 1 (PD-1) monoclonal antibody that binds to PD-1, aiming to restore the anti-tumor activity of T cells. Domvanalimab is an Fc-silent monoclonal antibody targeting the T-cell immunoreceptor with Ig and ITIM domains (TIGIT), a checkpoint receptor on immune cells that acts as a brake on anticancer immune responses. By blocking TIGIT, domvanalimab frees up immune pathways and activates immune cells to attack cancer without depleting regulatory T cells. The combination targets both PD-1 and TIGIT checkpoints, which play complementary roles in suppressing anti-tumor immunity, thereby enhancing overall immune activation against tumors. This regimen is being evaluated in multiple phase 3 clinical trials for various cancers including non-small cell lung cancer (NSCLC) and gastrointestinal cancers[4][5][6].
