Zinc metal-organic framework + 5-fluorouracil is an experimental drug delivery system designed to enhance the efficacy and reduce the systemic toxicity of the chemotherapeutic agent 5-fluorouracil (5-FU). The system utilizes a zinc-based metal-organic framework (Zn-MOF), a porous crystalline material, as a nanocarrier to encapsulate 5-FU. In advanced formulations, the Zn-MOF is functionalized with materials like folate-conjugated chitosan (FA-Cs) to enable pH-responsive controlled release and active targeting of folate receptors, which are frequently overexpressed in cancer cells such as hepatocellular carcinoma (HCC). Once delivered, 5-FU acts as a pyrimidine analogue that inhibits thymidylate synthase and incorporates into RNA and DNA, leading to cell cycle arrest and apoptosis.
