Overview

**ZKAB001 + capecitabine** is an investigational combination therapy composed of ZKAB001, an anti–PD-L1 monoclonal antibody, and capecitabine, an orally administered prodrug of 5-fluorouracil. ZKAB001 (also called socazolimab) is a human anti–PD-L1 antibody that blocks the interaction between PD-L1 and PD-1 receptors on T lymphocytes, enhancing anti-tumor immune responses. Capecitabine is a small molecule prodrug that is enzymatically converted into 5-fluorouracil in tumor cells, where it inhibits thymidylate synthase, DNA synthesis, and RNA function resulting in apoptosis of rapidly dividing malignant cells. The combination is being studied primarily in high-grade, localized osteosarcoma, leveraging capecitabine's antimetabolite chemotherapy effects with ZKAB001's immune checkpoint blockade to improve event-free survival and reduce recurrence.
