ZM008 + pembrolizumab is an investigational **combination therapy** composed of ZM008, a first-in-class fully human IgG1 monoclonal antibody targeting **LLT1 (CLEC2D)**, and **pembrolizumab**, an anti-PD-1 checkpoint inhibitor antibody. ZM008 disrupts the LLT1-CD161 interaction between natural killer (NK) cells and tumor cells, which is believed to facilitate tumor immune escape. By blocking this pathway, ZM008 activates NK cells, enhances immune cell infiltration, and promotes cytotoxic killing of tumor cells. Pembrolizumab blocks the PD-1 pathway on T cells, boosting adaptive immune responses against tumor cells. Preclinical and ex vivo studies showed that the combination more effectively reduced tumor growth and promoted immune cell infiltration in solid tumors, including non-small cell lung cancer (NSCLC) and muscle invasive bladder cancer (MIBC), compared to each monotherapy. The combination is currently under evaluation in a Phase 1 clinical trial for advanced solid tumors that are unresponsive to standard therapies.
