ZN-A-1041 + trastuzumab emtansine is an investigational oral combination therapy consisting of ZN-A-1041, a brain-permeable, selective HER2-targeted tyrosine kinase inhibitor, and trastuzumab emtansine (also known as T-DM1, or Kadcyla), an antibody-drug conjugate that combines trastuzumab (a monoclonal antibody targeting HER2) with the cytotoxic agent emtansine (DM1). ZN-A-1041 is designed for improved blood-brain barrier penetration to address HER2-positive cancers with brain metastases. Preclinical and early clinical data indicate additive or synergistic antitumor activity when these two agents are used together, especially in HER2-positive metastatic breast cancer, including cases with brain metastases. The primary mechanism involves dual HER2 inhibition by blocking receptor signaling (kinase inhibition and antibody-mediated) and targeted cytotoxic delivery to HER2-overexpressing tumor cells[3][7].
