Overview
ZN-c5 + palbociclib is an investigational combination therapy for advanced or metastatic estrogen receptor-positive (ER+), human epidermal growth factor receptor 2-negative (HER2-) breast cancer. ZN-c5 is a novel, orally bioavailable small-molecule selective estrogen receptor degrader (SERD) that promotes degradation of the estrogen receptor, thereby inhibiting ER-driven tumor growth. Palbociclib is a selective cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor that blocks cell cycle progression from G1 to S phase by preventing phosphorylation of the retinoblastoma protein. The combination aims to enhance anti-tumor efficacy by simultaneously targeting hormone signaling and cell cycle regulation pathways. Clinical trials are ongoing to evaluate safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with previously treated ER+/HER2- advanced breast cancer[1][3][4].
