Drug intelligence / Profile preview
The combination of zocilurtatug pelitecan (formerly ZL-1310) and obrixtamig (BI 764532) is an investigational dual-targeting therapeutic regimen designed to treat delta-like ligand 3 (DLL3)-expressing malignancies. Zocilurtatug pelitecan is a next-generation antibody-drug conjugate (ADC) consisting of a humanized anti-DLL3 monoclonal antibody conjugated to a novel camptothecin derivative (a topoisomerase 1 inhibitor) payload via a protease-cleavable linker using the TMALIN platform. Obrixtamig is an IgG-like bispecific T-cell engager (TCE) that simultaneously binds to DLL3 on tumor cells and CD3 on T-cells, redirecting the patient's immune system to selectively lyse DLL3-expressing cancer cells. This combination is being co-developed by Boehringer Ingelheim and Zai Lab under a clinical collaboration. It is currently being evaluated in the Phase Ib/II DAREON-36 clinical trial (NCT07707895) for the treatment of patients with advanced small cell lung cancer (SCLC) and other poorly differentiated neuroendocrine carcinomas (NECs), both as a doublet and as a triplet regimen in combination with the PD-L1 inhibitor atezolizumab.
