Overview
zofenopril + amlodipine is an oral fixed-dose combination of two antihypertensive agents: zofenopril, an angiotensin-converting enzyme inhibitor (ACEI), and amlodipine, a dihydropyridine calcium channel blocker. Zofenopril acts by inhibiting ACE, thereby reducing the conversion of angiotensin I to angiotensin II, leading to vasodilation, reduced blood pressure, and additional antioxidant and cardioprotective effects linked to increased hydrogen sulfide and nitric oxide bioavailability[2][4]. Amlodipine lowers blood pressure by selectively inhibiting calcium ion influx into vascular smooth muscle, causing arterial vasodilation and reducing peripheral resistance[5]. This combination is primarily indicated for the treatment of hypertension, particularly in patients inadequately controlled with monotherapy, and has shown additive benefits in reducing cardiovascular events after acute myocardial infarction[4].
