Zolbetuximab + mFOLFOX6 is a **combination therapy** consisting of **zolbetuximab**, a monoclonal antibody targeting claudin 18.2, and the **mFOLFOX6 chemotherapy regimen**, which includes oxaliplatin, leucovorin (folinic acid), and 5-fluorouracil. Zolbetuximab acts by binding to claudin 18.2 on tumor cells and inducing antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity, leading to cancer cell death. mFOLFOX6 is an established antineoplastic regimen; oxaliplatin forms DNA adducts causing cytotoxicity, 5-fluorouracil inhibits thymidylate synthase disrupting DNA synthesis, and leucovorin enhances the binding of 5-fluorouracil to thymidylate synthase. This combination is being investigated particularly for advanced and metastatic gastrointestinal cancers, such as gastric or gastroesophageal junction adenocarcinoma.
