Overview

Zoldonrasib + daraxonrasib is an investigational combination therapy developed by Revolution Medicines, consisting of two distinct RAS(ON) inhibitors. Zoldonrasib (RMC-9805) is an oral, tri-complex, covalent inhibitor that selectively targets the active (ON) state of the KRAS G12D mutation. Daraxonrasib (RMC-6236) is a multi-selective, noncovalent RAS(ON) inhibitor designed to inhibit a broad spectrum of RAS mutations. By combining a mutation-selective inhibitor with a multi-RAS inhibitor, the regimen aims to maximize therapeutic pressure on RAS signaling and prevent or overcome resistance in patients with KRAS G12D-mutant advanced solid tumors, including pancreatic ductal adenocarcinoma (PDAC) and non-small cell lung cancer (NSCLC).
