The combination of zoledronic acid and aromatase inhibitors is used in the treatment of hormone receptor-positive breast cancer, particularly in postmenopausal women. Zoledronic acid is a bisphosphonate that inhibits osteoclast-mediated bone resorption, while aromatase inhibitors (such as letrozole, anastrozole, or exemestane) block estrogen synthesis by inhibiting the enzyme aromatase. This combination has been shown to improve disease-free survival and reduce bone loss associated with endocrine therapy in breast cancer patients[1][3][5]. The sequence of administration may be important for maximizing anti-tumor effects; preclinical studies suggest that sequential treatment with an aromatase inhibitor followed by zoledronic acid may induce greater apoptosis than either drug alone or simultaneous administration[1].
