This inhibition prevents the prenylation of small GTP-binding proteins essential for osteoclast function and survival, thereby reducing bone resorption and increasing bone mineral density. Zoledronic acid is a more potent, third-generation bisphosphonate compared to the second-generation pamidronate, and is typically administered as a once-yearly infusion for osteoporosis, whereas pamidronate often requires more frequent dosing.
