Drug intelligence / Profile preview
Varicella zoster virus glycoprotein E adjuvanted with AS01B (commercially known as Shingrix) is a non-live, recombinant subunit vaccine developed by GSK for the prevention of herpes zoster (shingles) and its complications, such as postherpetic neuralgia. The vaccine consists of 50 μg of recombinant varicella-zoster virus glycoprotein E (gE) antigen, the most abundant surface protein of the virus, formulated with the liposome-based AS01B adjuvant system containing monophosphoryl lipid A (MPL) and the saponin QS-21. It works by strongly enhancing pre-existing gE-specific CD4+ T-cell and anti-gE antibody responses to overcome age-related decline in immunity. Shingrix was approved by the U.S. FDA in 2017 and is indicated for adults aged 50 years and older, as well as immunocompromised adults aged 18 years and older. Additionally, Leids Universitair Medisch Centrum (LUMC) is evaluating a fractional intradermal dose of the vaccine in a Phase 2-3 trial in the Netherlands to assess non-inferiority to the standard intramuscular dose.
