Overview
Shingrix is a non-live, recombinant subunit vaccine developed by GSK for the prevention of herpes zoster (shingles) and postherpetic neuralgia. It consists of the varicella-zoster virus (VZV) glycoprotein E (gE) antigen, which is produced in Chinese hamster ovary (CHO) cells, combined with the AS01B adjuvant system. The AS01B adjuvant system contains 3-O-desacyl-4'-monophosphoryl lipid A (MPL) and the saponin QS-21, which together enhance the cellular and humoral immune response to the gE antigen. This is particularly important for overcoming immunosenescence in older adults and providing protection to immunocompromised individuals. The vaccine was FDA-approved in 2017 and is administered as a two-dose intramuscular series.
