Drug intelligence / Profile preview
Zugo-cel + pirtobrutinib (zugocaptagene geleucel + pirtobrutinib) is an investigational combination therapy being evaluated for the treatment of aggressive B-cell lymphomas. The combination pairs zugocaptagene geleucel (zugo-cel, formerly CTX112), an off-the-shelf, allogeneic chimeric antigen receptor (CAR) T-cell therapy targeting CD19, with pirtobrutinib (formerly LOXO-305), an orally active, highly selective, non-covalent Bruton's tyrosine kinase (BTK) inhibitor. Zugo-cel is engineered using CRISPR-Cas9 technology to disrupt specific genes (such as TRAC, B2M, TGFBR2, and ZC3H12A) to enhance immune evasion, T-cell potency, and persistence without requiring HLA matching. Pirtobrutinib reversibly binds BTK, maintaining activity even in the presence of common covalent BTK inhibitor resistance mutations (such as C481 substitutions). CRISPR Therapeutics, in collaboration with Eli Lilly, is evaluating this combination in a Phase 1/2 clinical trial (NCT05643742) for patients with relapsed or refractory B-cell malignancies, aiming to achieve deep and durable clinical responses.
