**zunsemetinib + methotrexate** is a combination of two pharmaceutical agents developed for the treatment of inflammatory diseases such as rheumatoid arthritis. Zunsemetinib (also known as ATI-450) is an investigational small molecule inhibitor of the MAPK-activated protein kinase 2 (MK2) pathway, developed as a targeted anti-inflammatory therapy. Methotrexate is a well-established disease-modifying antirheumatic drug (DMARD) and folate antagonist that inhibits dihydrofolate reductase, thus impairing DNA/RNA synthesis and suppressing immune and inflammatory pathways. The combination is intended to provide synergistic benefit in diseases where monotherapy is suboptimal, particularly in rheumatoid arthritis.
