Overview

MenA, also known as 1,4-dihydroxy-2-naphthoate (DHNA) octaprenyltransferase, is a key enzyme in the biosynthetic pathway of menaquinone (vitamin K2) in bacteria. It catalyzes the penultimate step of menaquinone biosynthesis by transferring an isoprenoid side chain to DHNA, resulting in the formation of demethylmenaquinone (DMK), which is subsequently methylated to produce menaquinone. This enzyme is essential for bacterial electron transport and ATP generation under anaerobic conditions, making it an attractive antibacterial target. Inhibiting MenA can be bactericidal against Gram-positive and anaerobically respiring Gram-negative bacteria.
