Overview

26S proteasome non-ATPase regulatory subunit 6 (PSMD6) is a non-catalytic component of the 19S regulatory particle of the 26S proteasome, which is the primary machinery for ATP-dependent protein degradation in eukaryotic cells (UniProt, Q15008). PSMD6 is specifically located within the lid subcomplex of the 19S particle and is essential for the structural integrity and assembly of the proteasome, facilitating the recognition and unfolding of polyubiquitinated substrates (PubMed, 28655765). By controlling the degradation of regulatory proteins, PSMD6 influences critical cellular processes such as the cell cycle, signal transduction, and the stress response (NCBI Gene, 5712). In the context of oncology, PSMD6 is frequently upregulated in various malignancies, where it supports the high metabolic demands and proteotoxic stress tolerance of cancer cells (PubMed, 31435141). While current therapeutic strategies focus on inhibiting the 20S catalytic core (e.g., with bortezomib), PSMD6 and other 19S subunits are emerging as potential targets for next-generation proteasome inhibitors aimed at overcoming resistance (PubMed, 33064984). Targeting PSMD6 disrupts the proteasome's ability to process substrates, ultimately triggering apoptosis through the activation of the unfolded protein response and the stabilization of pro-apoptotic factors.
