Overview

Brassica juncea HMG-CoA synthase 1 (BjHMGS1) is a critical enzyme in the mevalonate (MVA) pathway of the Indian mustard plant, where it catalyzes the condensation of acetyl-CoA and acetoacetyl-CoA to form 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) (UniProt P54870). This enzyme serves as a key regulatory point for the biosynthesis of various isoprenoids, including phytosterols and brassinosteroids, which are essential for plant growth, development, and response to environmental stresses (Nagegowda et al., 2004, PubMed: 15107595). Research has demonstrated that overexpressing BjHMGS1, particularly the S359A mutant version, can significantly increase sterol content and enhance growth and stress tolerance in transgenic plants (Wang et al., 2012, PubMed: 22403514). While not a traditional human therapeutic target, BjHMGS1 is a major focus in agricultural biotechnology for improving crop yield, resilience, and nutritional value (Liao et al., 2014, PubMed: 24911014). The enzyme is sensitive to specific inhibitors such as hymeglusin (F-244), which are utilized in biochemical studies to probe metabolic flux within the MVA pathway (PubChem CID 6442177). Furthermore, BjHMGS1 expression is upregulated in response to abiotic stresses like chilling and wounding, highlighting its role in plant defense mechanisms.
