3-Phosphoinositide-dependent protein kinase-1 (PDK1) is a master serine/threonine kinase that plays a central role in the PI3K/Akt signaling pathway [2, 6]. It is responsible for the phosphorylation and activation of at least 23 members of the AGC kinase family, including Akt, S6K, SGK, and PKC [2, 10]. These downstream effectors regulate critical cellular processes such as growth, proliferation, survival, and metabolism [2, 4]. PDK1 is frequently overexpressed or hyperactivated in various human cancers, including breast, prostate, and pancreatic cancer, where it promotes tumor progression and metastasis [1, 4, 5]. Beyond oncology, PDK1 is involved in insulin signaling and glucose homeostasis, making it a potential target for metabolic disorders like type 2 diabetes [3, 7]. It has also been implicated in neurodegenerative diseases like Alzheimer's and cardiovascular conditions such as cardiac hypertrophy [7, 8]. While several small-molecule inhibitors have been developed to target its ATP-binding site or allosteric PIF pocket, none have yet reached FDA approval [9, 10, 22]. Therapeutic challenges include potential systemic toxicity and hyperglycemia due to its essential role in normal physiological functions [2, 11]. PDK1 remains a high-interest target for overcoming resistance to other PI3K pathway inhibitors in clinical settings [5, 8].
Inhibition of kinase activity by competing with ATP or binding to allosteric sites (PIF pocket), thereby preventing the phosphorylation and activation of downstream AGC kinases such as Akt, S6K, and PKC [2, 7, 10].
