Overview

39S ribosomal protein L4, mitochondrial (MRPL4) is a nuclear-encoded protein that forms part of the large subunit (39S) of the mitochondrial ribosome, which is responsible for protein synthesis within mitochondria. MRPL4, like other mitochondrial ribosomal proteins, differs substantially in sequence across species, making homology-based identification difficult. Mammalian mitoribosomes, composed of about 75% protein to rRNA, play a central role in synthesizing proteins critical for oxidative phosphorylation and cellular energy supply. MRPL4 has also been shown in animal models to regulate Notch signaling independently of its role in translation, affecting developmental processes. There is currently no evidence that MRPL4 is a direct therapeutic target, nor are there drugs or safety concerns associated with the protein in humans.
