Overview

5'-Nucleotidase domain-containing protein 3 (NT5DC3) is a 548-amino-acid, ~60–63 kDa protein with homology to nucleotidase enzymes. It is classified as an enzyme of the 5'-nucleotidase family capable of participating in hydrolase activity, although its specific substrate and molecular activity in vivo remain uncertain. Recent research highlights NT5DC3’s downregulation in breast and colon cancers, and its critical role as a tumor suppressor under hyperglycemic conditions. NT5DC3 acts as a biomarker for type 2 diabetes and the coexistence of diabetes and colon cancer, and its regulation—especially by the dietary agent lactoferrin—may be pivotal for prognosis and therapy monitoring. NT5DC3 is subject to complex epigenetic regulation via RNA and DNA methylation. Current data does not uncover any existing drugs targeting NT5DC3 directly, nor does it elucidate its full physiological function, but it remains a promising molecular target for future therapy research
