The 5T4 antigen (TPBG) is an oncofetal cell surface glycoprotein overexpressed across multiple solid tumors, including colorectal, ovarian, and gastric cancers. It is rarely found in healthy adult tissue, with limited expression during development or in certain neuronal subtypes. Its role includes promoting tumor progression through effects on cell movement and signaling pathways like Wnt. This makes it an attractive target for diverse anti-cancer therapies including vaccines, antibody-drug conjugates, superantigen-based approaches, and CAR-T cells.
