Overview

Abelson interactor 1 (ABI1) is a multi-domain adaptor protein that serves as a critical scaffold within the WAVE regulatory complex (WRC), facilitating actin cytoskeleton remodeling (UniProt P62495). By linking upstream signals from Rho-family GTPases like Rac1 to the Arp2/3 complex, ABI1 regulates essential cellular processes including motility, endocytosis, and cell-cell adhesion (PubMed: 10406804). In clinical contexts, ABI1 is notably involved in hematologic malignancies through the formation of ABI1-MLL fusion genes and is frequently overexpressed or dysregulated in various solid tumors, promoting invasive and metastatic phenotypes (PubMed: 25659580). Although direct pharmacological inhibitors of ABI1 are not yet in clinical use, it remains a target of interest due to its role in modulating ABL1 kinase activity and its influence on pathways targeted by tyrosine kinase inhibitors (PubMed: 15601771). Furthermore, its involvement in neuronal dendritic spine morphogenesis suggests potential implications in neurodevelopmental and cognitive disorders (PubMed: 29749331). As a key regulator of the actin machinery, ABI1 represents a potential node for therapeutic intervention in diseases characterized by aberrant cell migration and invasion.
