The Staphylococcus aureus agr quorum-sensing system is a sophisticated regulatory network essential for controlling virulence gene expression based on population density. It enables rapid adaptation between colonization/biofilm states and aggressive tissue invasion through coordinated regulation mediated primarily by autoinducing peptides sensed via a two-component signaling module (AgrC/AgrA) with downstream effects executed largely through the multifunctional regulatory sRNA RNAIII.
Inhibition of AgrC histidine kinase activity, disruption of AgrA phosphorylation, interference with AIP binding, or targeting RNAIII function to block downstream gene expression. Some drugs aim to prevent AIP processing or secretion.
