Activation of helper CD4+ T-cells refers to the process by which these immune cells, also known as T helper (Th) cells or CD4+ T-cells, become functionally active in response to antigenic stimulation. These cells are central coordinators of the adaptive immune response and play a critical role in orchestrating both cellular and humoral immunity. The activation process is highly regulated and requires three distinct signals: antigen recognition, costimulation, and cytokine signaling. Upon activation, naive CD4+ cells differentiate into specialized subsets such as Th1 or Th2 based on cytokine cues, enabling them to orchestrate defense against intracellular pathogens or extracellular parasites, respectively. Dysregulation can lead to autoimmunity due to inappropriate breaking of tolerance mechanisms.
