Overview

Adhesion G protein-coupled receptor E4 (ADGRE4), also frequently referred to as FIRE (F4/80-like receptor) or EMR4, is a member of the EGF-TM7 subfamily within the adhesion G protein-coupled receptor (aGPCR) family. It is predominantly expressed on the surface of myeloid cells, including specific subsets of macrophages and dendritic cells, and is known to facilitate cellular interactions between these antigen-presenting cells and B lymphocytes. In humans, the ADGRE4 gene is generally classified as a pseudogene (ADGRE4P) due to a specific frameshift mutation that typically results in a truncated, soluble protein rather than a functional seven-transmembrane receptor. However, research indicates potential expression or relevance in human disease states like glioblastoma and inflammatory disorders, suggesting it may still serve as a biomarker or specialized mediator. Despite its orphan receptor status and pseudogene classification in humans, it remains a valuable target for investigating myeloid cell biology and the evolutionary divergence of immune signaling pathways.
