Adiponectin, C1Q and collagen domain containing (ADIPOQ) is a secreted protein produced exclusively by adipose tissue. It circulates in plasma and structurally resembles collagens and complement factor C1q. Adiponectin is a multifunctional adipokine regulating glucose and lipid metabolism, improving insulin sensitivity, and exerting anti-inflammatory, anti-diabetic, and anti-atherogenic effects. It activates AMP-activated protein kinase (AMPK) pathways in muscle and liver, enhances glucose and fatty acid usage, and antagonizes inflammatory cytokines such as TNF-alpha. Clinical interest in adiponectin centers on its role in metabolic diseases, cardiovascular disease, and as a biomarker for insulin resistance and risk assessment.
Drugs increasing adiponectin levels act via enhanced gene expression and secretion in adipocytes. Drugs may act through agonism of the PPARγ receptor, resulting in upregulated adiponectin synthesis and secretion.
