Due to their widespread distribution, selectivity is a key pharmacological consideration to avoid adverse effects like bronchoconstriction or metabolic interference [StatPearls: NBK534230]. Overall, the adrenoceptor system is a fundamental component of the autonomic nervous system, regulating critical homeostatic functions across the cardiovascular, respiratory, and metabolic systems.
Drugs targeting adrenoceptors act as either agonists or antagonists to modulate the sympathetic nervous system. Agonists mimic the action of endogenous catecholamines to activate G protein-mediated signaling pathways: alpha-1 agonists trigger the Gq-PLC-IP3 pathway leading to calcium release and muscle contraction, while beta agonists activate the Gs-adenylyl cyclase-cAMP pathway to stimulate cardiac activity or relax smooth muscle [StatPearls: NBK534230]. Antagonists, such as beta-blockers or alpha-blockers, competitively bind to these receptors to inhibit the effects of sympathetic stimulation, thereby reducing blood pressure, heart rate, or smooth muscle tone [IUPHAR: Adrenoceptors].
