Overview

Alpha-latrotoxin is a potent 130 kDa neurotoxin found in the venom of the Southern black widow spider, Latrodectus mactans (UniProt: P08037). It specifically targets vertebrate presynaptic nerve terminals, where it binds with high affinity to receptors such as Latrophilin-1 and Neurexin-1-alpha (PubMed: 11752545). Upon binding, the toxin inserts into the presynaptic membrane to form non-selective cation pores, which allow for a massive influx of calcium ions (PubMed: 18411774). This influx, along with receptor-mediated signaling, triggers the exhaustive and uncontrolled release of neurotransmitters like acetylcholine and glutamate. The resulting clinical condition, known as latrodectism, is characterized by severe muscle pain, abdominal rigidity, and autonomic instability (StatPearls: NBK430844). In a research context, alpha-latrotoxin is a critical pharmacological tool used to investigate the molecular mechanisms of synaptic vesicle exocytosis. From a therapeutic perspective, the toxin is the primary target for specific antivenoms designed to neutralize its activity and alleviate symptoms of envenomation. Safety concerns regarding this molecule primarily involve its extreme neurotoxicity and the potential for life-threatening complications like respiratory failure or hypertensive crisis.
