Amelogenin, Y isoform (AMELY) is a gene located on the Y chromosome that encodes a form of the amelogenin protein, which is an extracellular matrix protein involved in the biomineralization process during tooth enamel development. Amelogenins form a family of proteins essential for the formation, organization, and mineralization of enamel but the vast majority of amelogenin is produced from the X-linked gene (AMELX); the Y isoform (AMELY) makes only a minor contribution and is not essential for normal enamel formation. AMELY is routinely used in molecular and forensic applications as a sex-determination marker due to its presence only in males. Unlike some protein targets, AMELY is not classified as a receptor, enzyme, ion channel, transporter, or transcription factor. There is no evidence that drugs directly interact with or target AMELY, nor does it play a central role in disease processes, although it is linked to the amelogenin gene family whose dysfunction (AMELX) is responsible for X-linked amelogenesis imperfecta.
