Overview

The Amiloride-sensitive sodium channel subunit delta (SCNN1D) is a critical component of the epithelial sodium channel (ENaC) complex, which facilitates the rate-limiting step of sodium reabsorption across epithelial membranes (UniProt P51170). Unlike the alpha, beta, and gamma subunits that are primarily found in the kidney, the delta subunit is highly expressed in the human brain, lung, and heart (NCBI Gene 6339). In the lungs, SCNN1D plays a vital role in maintaining the volume of airway surface liquid, which is essential for effective mucociliary clearance and respiratory health (PubMed 32824143). Dysfunction or overactivity of this channel is linked to respiratory conditions such as bronchiectasis and cystic fibrosis-like lung disease (PubMed 32824143). The channel is characterized by its high sensitivity to the diuretic drug amiloride, which binds to the extracellular loop and blocks the ion-conducting pore (PubChem CID 2162). Because of its unique tissue distribution, SCNN1D is considered a potential target for treating pulmonary and neurological disorders without the profound renal effects associated with other ENaC subunits (PubMed 22461022). Pharmacological modulation of SCNN1D aims to balance sodium levels and fluid homeostasis to alleviate symptoms of fluid overload or impaired clearance (UniProt P51170).
