Overview

Amyloid-beta (Aβ) is a peptide derived from the proteolytic cleavage of the Amyloid Precursor Protein (APP) and is the primary component of senile plaques in Alzheimer's disease (UniProt P05067). The N-terminal epitope, specifically residues 1-9 (sequence: DAEFRHDSG), represents a highly immunogenic region that is solvent-exposed in both soluble oligomers and insoluble fibrils (PubMed: 11160536). This specific epitope is a major target for passive and active immunotherapy, as antibodies binding to the N-terminus can trigger the clearance of Aβ deposits through microglial-mediated phagocytosis (PubMed: 20147467). Several clinical-stage monoclonal antibodies, such as Bapineuzumab and Gantenerumab, recognize sequences within or overlapping this N-terminal segment to reduce amyloid burden in the brain (PubMed: 22431022). Beyond its role in pathology, the N-terminus of Aβ is known to bind divalent metal ions like copper and zinc, which may influence its aggregation state and oxidative stress levels (PubMed: 12031246). Therapeutic targeting of this region must be carefully managed due to the risk of Amyloid-Related Imaging Abnormalities (ARIA), which result from the removal of amyloid from the cerebrovascular walls (PubMed: 28231025).
