Overview

Angiopoietin-2 (ANGPT2) is a secreted glycoprotein that serves as a key regulator of vascular remodeling and angiogenesis (UniProt: O15123). It primarily acts as a context-dependent antagonist of the Tie2 receptor tyrosine kinase, competing with the stabilizing ligand Angiopoietin-1 (ANGPT1) to promote vascular destabilization, which is necessary for vessel sprouting in the presence of VEGF (PubMed: 28611162). In pathological conditions such as cancer and neovascular eye diseases, ANGPT2 is frequently overexpressed, contributing to abnormal vessel growth, increased vascular permeability, and inflammation (PubMed: 31439490). Therapeutic strategies targeting ANGPT2, such as the bispecific antibody faricimab, aim to stabilize the vasculature and reduce pathological leakage by blocking ANGPT2-Tie2 signaling (DrugBank: DB15055). Additionally, ANGPT2 inhibition is being explored in oncology to normalize tumor vasculature and enhance the efficacy of other therapies (PubMed: 24501218).
