Overview

Angiotensin I is a decapeptide (Asp-Arg-Val-Tyr-Ile-His-Pro-Phe-His-Leu) that serves as a critical intermediate in the renin-angiotensin system (RAS), a key hormonal cascade for regulating blood pressure and fluid balance [1][2]. It is produced when the enzyme renin cleaves its precursor, angiotensinogen, which is primarily synthesized in the liver [3]. While Angiotensin I itself is biologically inactive or possesses very limited activity, it is rapidly converted into the potent vasoconstrictor Angiotensin II by the action of angiotensin-converting enzyme (ACE), primarily in the pulmonary circulation [3][4]. Because of its central role as a substrate in this pathway, Angiotensin I levels are frequently measured to assess the activity of the RAS and the efficacy of antihypertensive medications [4]. Drugs such as ACE inhibitors and renin inhibitors modulate the levels of Angiotensin I to treat conditions like hypertension, heart failure, and chronic kidney disease [3]. Consequently, while not a direct therapeutic target itself, Angiotensin I is a vital biomarker and metabolic pivot point in cardiovascular medicine [1][2].
