Overview

Anion exchange protein 1 (AE1), commonly known as Band 3, is the most abundant integral membrane protein in erythrocytes, comprising approximately 25% of the total membrane protein content (UniProt: P02730). It functions primarily as a chloride/bicarbonate exchanger, facilitating the transport of CO2 from tissues to the lungs, and serves as a crucial structural scaffold by anchoring the plasma membrane to the spectrin-based cytoskeleton via interactions with ankyrin and protein 4.2 (PubMed: 21810361). Mutations in the SLC4A1 gene are linked to several disorders, including hereditary spherocytosis, Southeast Asian ovalocytosis, and distal renal tubular acidosis (NIH Gene: 6521). Furthermore, Band 3 is a key entry receptor for Plasmodium falciparum, the parasite responsible for malaria, making it a target for potential anti-malarial interventions (PubMed: 15548731). Although direct pharmacological targeting of Band 3 is currently limited to research tools like stilbene disulfonates (e.g., DIDS), its role in maintaining RBC integrity and acid-base balance makes it a significant focus for hematological and physiological research.
