Annexin A8 (ANXA8) is a member of the annexin family, a group of evolutionarily conserved proteins characterized by their ability to bind phospholipids in a calcium-dependent manner (UniProt P13927). ANXA8 is specifically involved in regulating membrane-cytoskeleton interactions and the trafficking of various cell surface receptors, such as CD133, which is a marker for cancer stem cells (PubMed: 20530696). In clinical contexts, ANXA8 is frequently upregulated in aggressive malignancies, including triple-negative breast cancer and pancreatic ductal adenocarcinoma, where it contributes to tumor progression, epithelial-mesenchymal transition (EMT), and resistance to apoptosis (PubMed: 27447547). While no small molecule inhibitors are currently in clinical use, the ANXA8 mRNA is a significant target for experimental RNA interference (RNAi) therapies designed to silence its expression and inhibit oncogenic signaling pathways (PubMed: 31232193). Understanding the precise role of ANXA8 in membrane dynamics and its differential expression in diseased versus healthy tissue remains a key focus for developing targeted therapeutic strategies.
