Overview

B-cell antigen receptors (BCRs) and T-cell receptors (TCRs) recognizing fluorescein isothiocyanate (FITC) epitopes are the primary components of the adaptive immune system targeted in hapten-directed immunotherapy (Leamon and Low, 2001). These receptors are typically induced through vaccination with a hapten-carrier conjugate, such as FITC-labeled Keyhole Limpet Hemocyanin (KLH), which stimulates the production of high-affinity anti-FITC antibodies and T-cells (Amato et al., 2013). Once a robust immune repertoire is established, a bispecific adapter molecule—comprising a targeting ligand such as folate linked to the FITC hapten—is administered to the patient (Amato et al., 2014). This adapter bridges the pre-existing anti-FITC receptors to cells overexpressing the target ligand's receptor, such as folate receptor-alpha on cancer cells or folate receptor-beta on activated macrophages (Paulos et al., 2004). The resulting recruitment of anti-FITC BCRs and TCRs to the disease site triggers potent immune effector functions, including antibody-dependent cellular cytotoxicity (ADCC) and direct T-cell-mediated lysis (ClinicalTrials.gov NCT00492167). This approach, notably developed as the FolateImmune platform, aims to treat various cancers and inflammatory conditions by redirecting the body's immune memory toward specific pathological cells.
