This target refers to the modulation of antigen presentation by controlling the expression of antigens encoded on delivered mRNA constructs, where this expression is regulated in a tissue-specific manner by endogenous or engineered miRNAs. By incorporating miRNA recognition elements into the 3′ UTR of an mRNA construct, its translation can be selectively repressed in tissues where a given miRNA is highly expressed, allowing for targeted antigen production in desired cell types like antigen-presenting cells (APCs). This approach enables precise control over which cell types present antigens and activate T cells, offering a flexible immunomodulation strategy.
