Caspase-8 is a key initiator caspase in the extrinsic apoptosis pathway, triggered by extracellular death signals binding to cell surface death receptors. It exists as an inactive proenzyme (zymogen) and requires proteolytic cleavage for activation. Activated caspase-8 cleaves and activates effector caspases leading to cellular disassembly and apoptosis. It can also cleave BID, linking extrinsic and intrinsic apoptotic pathways. Dysfunction in caspase-8 activity has been linked with developmental abnormalities, immune system disorders and cancer progression.
