Arabinogalactan endo-beta-1,4-galactanase (EC 3.2.1.89) is a glycoside hydrolase, primarily classified within the GH53 family, that catalyzes the internal hydrolysis of (1->4)-beta-D-galactosidic linkages in type I arabinogalactans [1, 6]. These polysaccharides are structural components of the "hairy" regions of pectin in plant cell walls, and their degradation is essential for biomass turnover and carbon acquisition by microorganisms [11, 13]. The enzyme is widely distributed among bacteria and fungi, including industrial strains like Aspergillus niger and human gut commensals such as Bacteroides thetaiotaomicron [3, 34]. In the human gut, it plays a crucial role in the metabolism of dietary galactooligosaccharides (GOS), which are recognized prebiotics that promote the growth of beneficial bifidobacteria [27, 34]. While not a traditional therapeutic target for human drugs, the enzyme is of significant interest in biotechnology for biofuel production, food processing, and the development of synbiotics to modulate the intestinal microbiome [11, 31]. Its activity is also relevant in plant pathology, where it contributes to the degradation of host cell walls by fungal and bacterial pathogens [26]. Research into its structure and function has facilitated the engineering of thermostable variants for industrial applications and the design of prebiotic mixtures for human health [27, 32]. There are currently no clinical drugs that target this enzyme, although it is used as a tool in biochemical research and diagnostic assays [1, 13].
