Overview

Arf-GAP with GTPase, ankyrin repeat, and PH domain-containing protein 3 (AGAP3) is a multi-domain synaptic signaling protein. It is a component of the NMDA receptor complex, where it functions as a GTPase-activating protein (GAP) for Arf6 and possesses a GTPase-like domain. AGAP3 links NMDA receptor activation to trafficking of AMPA-type glutamate receptors during long-term potentiation (LTP), a fundamental mechanism of synaptic plasticity and memory. AGAP3 contains an N-terminal GTPase-like domain, a pleckstrin homology (PH) domain that mediates protein or membrane interactions, an ArfGAP domain involved in GTPase activation of ADP-ribosylation factors, and C-terminal ankyrin repeats. AGAP3 regulates Ras/ERK and Arf6 signaling cascades and is essential for proper AMPA receptor surface expression and synaptic function. Its dysregulation may impact memory consolidation and cognitive plasticity, but it is not currently an established direct therapeutic target[1][2][3].
