Overview

Arrestin beta-2 (ARRB2), also known as Beta-arrestin-2, is a ubiquitous intracellular scaffolding and adaptor protein that plays a fundamental role in regulating G protein-coupled receptor (GPCR) signaling (UniProt P32121). It is primarily responsible for the desensitization and internalization of activated GPCRs, effectively dampening G protein-mediated responses while simultaneously initiating independent signaling cascades, such as the MAPK and Akt pathways (PubMed 11261574). ARRB2 is implicated in a wide range of pathological conditions, including chronic pain, cardiovascular diseases, and various cancers, where it can act as either a promoter or suppressor of tumor progression (PubMed 2901574, MDPI 1.4.5). Therapeutic strategies targeting ARRB2 include the development of biased GPCR agonists that selectively avoid or recruit arrestins, as well as RNA-targeted approaches like siRNA and antisense oligonucleotides designed to modulate ARRB2 mRNA levels (NIH 1.1.2, 1.4.1). Given its central role in cellular homeostasis, targeting ARRB2 requires careful consideration of its diverse functions across different tissues to avoid significant off-target effects (ResearchGate 1.4.2). Research has also highlighted its role in immune checkpoint regulation, making it a potential target for enhancing immunotherapy in diseases like multiple myeloma (MDPI 1.4.1).

Mechanism of action

Drugs targeting ARRB2 mRNA utilize RNA interference (siRNA) or antisense oligonucleotides (ASOs) to induce the degradation of the transcript, thereby reducing the expression of the Arrestin beta-2 protein (NIH.gov). Other approaches include small molecules like Barbadin that inhibit protein-protein interactions between ARRB2 and endocytic machinery, or biased GPCR agonists that indirectly modulate ARRB2 recruitment to receptors (PubMed 23045611).
